Read the <a href=\"https:\/\/theconversation.com\/a-promising-new-approach-to-treating-potentially-deadly-liver-disease-253924\" target=\"_blank\" rel=\"noreferrer noopener\">original article<\/a>.<\/h6>\n\n\n\n<p><a href=\"https:\/\/theconversation.com\/profiles\/maria-teresa-borrello-2364636\" target=\"_blank\" rel=\"noreferrer noopener\">Maria Teresa Borrello<\/a>, <em><a href=\"https:\/\/theconversation.com\/institutions\/university-of-sunderland-1223\" target=\"_blank\" rel=\"noreferrer noopener\">University of Sunderland<\/a><\/em><\/p>\n\n\n\n<p class=\"has-large-font-size\">An experimental new treatment is showing early promise in the fight against liver fibrosis \u2013 a serious and often silent condition that affects around <a href=\"https:\/\/commonslibrary.parliament.uk\/research-briefings\/cdp-2024-0070\/\" target=\"_blank\" rel=\"noreferrer noopener\">two million people<\/a> in the UK.<\/p>\n\n\n\n<p>Liver fibrosis happens when the liver <a href=\"https:\/\/pubmed.ncbi.nlm.nih.gov\/26159264\/\" target=\"_blank\" rel=\"noreferrer noopener\">becomes damaged<\/a> \u2013 often due to long-term issues like alcohol use, obesity or chronic infections \u2013 and starts to develop scar tissue. Over time, that scarring can get worse and lead to serious complications such as liver failure or cancer.<\/p>\n\n\n\n<p>The problem is that most people don\u2019t know they have it until the damage is advanced. Over time, this repair process can become unbalanced, leading to a buildup of scar tissue.<\/p>\n\n\n\n<p>A key part of this process involves hepatic stellate cells. Our recent research suggests it also plays an important role in turning on the liver cells that cause scarring after injury. That\u2019s why we\u2019re exploring HDAC6 as a potential target for new treatments that could help prevent or even reverse liver fibrosis.<\/p>\n\n\n\n<p>In our lab, we developed two new drugs specifically designed to block HDAC6 activity. <\/p>\n\n\n\n<h2 class=\"wp-block-heading\">Liver slices<\/h2>\n\n\n\n<p>To see if these compounds could be useful as treatments, we tested them on precision-cut slices of human liver tissue at Newcastle University. This model keeps the <a href=\"https:\/\/pubmed.ncbi.nlm.nih.gov\/30963615\/\" target=\"_blank\" rel=\"noreferrer noopener\">liver\u2019s natural 3D structure and mix of cells<\/a>, making it a valuable way to study how diseases develop and how drugs might work.<\/p>\n\n\n\n<p>Our results were striking. Treating the liver slices with HDAC6 inhibitors greatly reduced signs of fibrosis, showing that these compounds can stop \u2013 and possibly even reverse \u2013 the scarring process at the cellular level.<\/p>\n\n\n\n<p>The inhibitors showed very little toxicity, suggesting they could be safe for further development.<\/p>\n\n\n\n<p>This research is a step forward in finding a treatment for liver fibrosis. Unlike previous treatments that targeted broad mechanisms or caused <a href=\"https:\/\/pubmed.ncbi.nlm.nih.gov\/35393658\/\" target=\"_blank\" rel=\"noreferrer noopener\">side-effects<\/a>, our HDAC6 inhibitors provide a more targeted approach. By focusing on a key cause of fibrosis, we may be able to stop the disease before it reaches irreversible stages.<\/p>\n\n\n\n<p>The implications are enormous. Liver disease is responsible for around 4 per cent of premature <a href=\"https:\/\/www.journal-of-hepatology.eu\/article\/S0168-8278(23)00194-0\/fulltext\" target=\"_blank\" rel=\"noreferrer noopener\">deaths globally<\/a>, and the burden is rising in line with alcohol misuse, obesity, and the use of multiple medications (known as \u201cpolypharmacy\u201d). A targeted therapy that interrupts fibrosis at its root could change the lives of tens of thousands of patients annually \u2013 not only in the UK but around the world.<\/p>\n\n\n\n<p>While these early findings are encouraging, more work is needed before HDAC6 inhibitors can be tested in humans.<\/p>\n\n\n\n<p>Our next steps include refining the experimental drugs, testing their effects in lab animals, and looking at how they might work alongside existing treatments.<\/p>\n\n\n\n<p>As researchers and healthcare professionals seek new ways to tackle chronic diseases, targeted approaches like this one could redefine how we treat conditions once considered untreatable. For patients with liver fibrosis, this new knowledge could mean a longer, healthier life for millions of people with liver fibrosis.<\/p>\n","protected":false},"excerpt":{"rendered":"<p>An
